چکیده

Background For rapid recombinant protein production in small to medium size volumes, transient transfection of mammalian cells is still the method of choice in biotechnology [1]. However, due to the high costs of commercially available lipofectamines or polycationic transfection reagents such as polyethylenimine (PEI), the most widely used transfection reagents available present a substantial economic bottleneck. While these reagents produce seemingly high transient transfection rates [2], there is still a strong desire for transfection reagents providing both secure and easy handling and higher recombinant protein production. As part of our commitment to excellence, InVivo BioTech Services initiated a joint venture with emp Biotech and developed a novel polycationic reagent, named INVect, for transient transfection and recombinant protein production in mammalian cells.

Reconciling pillars of transient gene expression: From DNA prep via media, reagent and cell line development to holistic process optimization

Background Transient gene expression (TGE) is a multi-parametric process which is built upon four essential influencing pillars: First of all, it is imperative to have an easy to transfect cell line which allows high product titers and is cultivated in suspension [1]. Secondly, only a few of the commercially available cell culture media allow/support transient transfection and production. Thus,...

Using transient transfected Chinese hamster ovary (CHO) cells by pET28a-LIC-NTF4 in gene therapy

Introduction: Neurotrophins, as a family of proteins, are responsible for induction of the survival, development, and function of neurons. Also, neurotropic factors are growth factors like Neurotrophins that help neurons survive. Moreover, Neurotrophins differentiate between progenitor cells so that neurons are formed. Despite the fact that the majority of mammalian brain neurons are produced p...

Polycationic lipids translocate lipopolysaccharide into HeLa cells.

We have investigated the ability of LIPOFECTAMINE, a polycationic lipid reagent used in DNA transfection, to translocate E. coli lipopolysaccharide (LPS) into HeLa cells. Although HeLa cells did not spontaneously take up fluorescein isothiocyanate-labelled LPS (FITC-LPS) from the culture medium, the cells that were co-incubated with greater than 1 g/mL FITC-LPS and LIPOFECTAMINE showed punctate...

Optimization of SW480 Colon Cancer Cells Transfection with Lipofectamine 2000

ABSTRACT           Background and Objectives: Nonviral carriers including those based on synthetic cationic lipids, offer several advantages over the viral counterparts. These carriers are able to form complexes with nucleic acids and deliver genes into the cells via the cellular endocytosis pathway, without significant toxicity. The level of transg...

Gene Delivery to Mesenchymal Stem Cells

There is increasing trend in using recombinant stem cells as novel therapeutic candidates in different diseases. These studies encompass different applications from targeted homing of Mesenchymal Stromal (stem) Cells (MSC), to arming them with different cytokines. Resistance to transfection or transduction methods had urged researchers to look for better gene delivery alternates and optimizing ...
